how hard will it be to go from the single tier bitnami in a VM to multi-tier bitnami in a VM (on AWS)?
It will depend on the type of project you are trying to address. I can't really provide you with a quantitative scale on how difficult it might be. Deploying a new site should be a simple process. Regular operations like connecting to an instance or managing certain components may require an understanding of the underlying architecture. In a Single Tier deployment, all components are located in the same server and these type of process are typically less complicated.
Does multi tier LAMP site behind a load balancer? I have modified my code to use mysql RDS so that a user can come in through any of the servers. Must I use a local copy of mysql when using multi-tier?

For the laptop install, should I be using a VM? Will this make it easier to transfer up to the multi tier on AWS or should I just be using Single tier?
A Single Tier installation on your laptop can be achieved by means of a SingleVM (all components are deployed on the machine) or a regular installer. In the Bitnami site, "Single Tier" assets is a term reserved for cloud instances, so you can't really install them in your computer.
